STANDARD OPERATING PROCEDURE FOR GCMS

Steps to be followed Switch on the instrument

Open the Helium gas cylinder (The outlet pressure of the gas should be about 50-60 psi), Switch on the GC followed by MS. Wait for 2 minutes. Switch on the PC

Double click the GCMS Real time Analysis icon GCMS Real time Analysis window opens, On the left side of the window there is a Assistant Bar, you click the Vaccum control icon (If it is not present than click top icon)

Once we click vaccum control icon, vaccum control window opens, just click the Auto start up.

Wait till the Vaccum created. Once Vaccum created go back to Data acquisition window by clicking the Data acquisition icon in Assistant bar.

Set the GC and MS parameter in the Data acquisition window. Than save the method file. Then Click the Acquisition icon followed by Download initial parameters. Wait for few minutes till instrument downloads all parameters.

TUNINIG 12 If the instrument is in continuous running condition we have to do tuning after 23 days interval. And if the instrument is shutdown than after restarting we have to do the tuning every time. 13 14 15 Click on the Tuning icon present in the assistant bar Click start auto tuning, Save the tuning file. After completion of tuning, click the View Tuning results icon to see the tuning results. (Check the FMHW parameter it should be in the range of 0.5 to 0.7. And the Peak of m/z 69 should be twice than m/z 28 peak.)

Sample Analysis 16 After setting GC and MS parameters save the method file. Single sample analysis 17 18 Click on the sample Login icon, sample login window open Give the name of the file, sample, select the latest tuning file, enter vial number, injection volume, than click OK. 19 Click on the Download parameter. Run will start automatically. Batch Analysis 20 Click the Batch processing icon, batch processing table open, enter all parameters, Save the batch file and click the start button.

View the result of the sample 21 You can check the chromatograms during Run by clicking the snapshot icon at Realtime Analysis window or Just go to GCMS Postrun analysis window and open the previous data files. 22 To check the mass spectra of the particular peak, click on the peak you will get the mass spectra. 23 To search for the library, click the required peak and than click the Similarty search. You will get the library search result for the particular peaks. 24 To change the library goes to Qualitative, Click the similarity search parameter, set the desired libraries. And click Ok. 25 To identify all the peaks automatically click the Integrate all the Peaks, give the integration parameters, 26 Click the Qualitative table, Than click TIC, Click edit, select all, followed by Registered to spectrum process table. 27 Click the Spectrum process table, Click on similarity search, search for all table, you will get the library search result for all the integrated peaks.

Shut Down the instrument 28 Click on Vaccum control icon, followed by Auto Shut down. Wait till auto shut down process completed. 29 Shut down the MS, GC followed by PC and Gas.
