Gram staining or the gram method is one of the most important and
broad staining techniques used to identify bacteria. Bacteria stained by
this method are divided in two groups, namely gram positive bacteria
and gram negative bacteria. Gram positive Bacteria will retain the
violet crystalline dye and will appear dark purple under a microscope.
The gram negative bacteria will lose the violet crystal dye after
washing with alcohol, and when given a coloring agent that is equal to
the water coloring agent fuchsine or safranin will appear red.
