a. Department where thesis work Medical Laboratory Technology,
will be carried out DDU Kaushal Kendra, RGSC,
Barkachha,
Banaras Hindu University, Varanasi,
Uttar Pradesh
b. Which other Department/ Faculty/ Departments providing access to
Institute will cooperate in the instrumentation such as FTIR, SDS-
proposed thesis PAGE, SEM, and DSC as required.
6.
a. Objectives of Proposed Work 1. To extract collagen from chicken
skin using acid hydrolysis.
2. To characterize extracted
collagen through techniques
including FTIR, SDS-PAGE, thermal
analysis, and SEM.
3. To evaluate the extracted
collagen’s suitability for biomedical
applications like tissue engineering
and wound healing.
b. Scope of Proposed Research Work The study explores chicken skin, an
underutilized poultry byproduct, as a
sustainable and cost-effective
collagen source. It aims to reduce
dependency on bovine/porcine
tissues, addressing issues like
zoonotic risks and religious
restrictions. This work ensures
biomedical viability through
structural and functional stability
assessments, with potential eco-
friendly and economic impacts.
7.
Lacunae in Knowledge on Subject Limited research focuses on
characterizing chicken skin-derived
collagen for its biomedical
properties. There is also insufficient
data comparing it to conventional
sources like bovine/porcine skin
collagen in terms of structural,
thermal, and morphological aspects.
8.
Preliminary Work, Which Has Preliminary trials on chicken skin
Already Been Done collection, defatting, and pre-
treatment with NaOH have
demonstrated the feasibility of
collagen extraction using acid
hydrolysis. Early characterization
trials confirm collagen's triple-helical
structure integrity.
9.
Whether Any Work Pertaining to Yes, studies have reported collagen
Proposed Work Has Been Done extraction from fish and poultry
Earlier byproducts; however, these lack a
comprehensive evaluation of the
biomedical potential of chicken skin
collagen.

Details of Procedure and Methods 1. Sample Preparation: Collection of
Proposed to Be Used in Study fresh chicken skin from local poultry,
followed by defatting and NaOH pre-
treatment.
2. Collagen Extraction: Acid
hydrolysis using 0.5M acetic acid
and subsequent centrifugation,
dialysis, and freeze-drying.
3. Characterization Techniques:
- FTIR Spectroscopy: To confirm
collagen-specific functional groups.
- SDS-PAGE: To assess molecular
weight and purity.
- Thermal Analysis (DSC): To
measure thermal stability.
- SEM Imaging: To observe fibrillar
structure and morphology.
4. Yield Calculation: Based on raw
skin weight and freeze-dried
collagen yield.
12.
Is Work Associated with Animal/ No animal/human subject
Human? involvement. The study uses
discarded chicken skin from poultry
waste.
