IV. PROCEDURE
Homemade Agar Plate Procedure
1. Prepare the ingredients needed for the homemade agar.
2. Sterilize all the equipment that are needed including your hands.
3. Pour four (4) cups of cold water together with the four (4) packs of gelatin in a
saucepan. Then, stir in eight (8) teaspoons of sugar and four (4) broth cubes (e.g.
knor cubes beef).
4. Then, heat the saucepan at medium low heat while continuously stirring it, and
wait until the mixture boils thoroughly then turn off the heat.
5. After turning off the heat, cool down the mixture for about 3-5 minutes.
6. Then, you can now fill the sterilized petri dishes/ microwavable plastic container
with the mixture for about 1/3.
7. Then, after using it all up you can proceed in covering the dishes with plastic
wrap or lid.
8. Lastly, you can now place it in the refrigerator for about 2-3 hour to allow the
mixture to cool and firm. If the homemade agar is ready, we can proceed to the
experiment.
Experiment procedure:
9. Ready five agar plates for this experiment, and label each one with its
corresponding specimen. Set one agar plate and label it with “control” using the
marker. Same procedure in other specimen. Remember not to open the dish until
the specimen is ready in order to avoid contamination.
10. For other four other agar plates (except for the “control”), identify four different
surfaces to test for bacteria. Utilize a cotton swab to collect bacteria samples.
Remember, only one surface per cotton swab.
11. After swabbing a surface, softly draw squiggly line across the agar and close the
lid. Use only one swab per agar plate.
Analyze your petri dishes every day and write down the observations in your
notebook

ANSWER: The microscope has the advantage of producing higher resolution than the naked
eye, which means that the higher the resolution, the better the ability to detect two or more
distinct objects without image blur.

ANSWER: The digestive tract health, reduction risk of heart disease, blood glucose control,
and weight management are all benefits of a high-fiber diet. Dietary fiber is a component of the
digestive tract that aids in the movement of food through the intestinal tract and the excretion of
solid waste. Soluble fiber binds with cholesterol-containing compounds in bile, resulting in a
cholesterol-lowering effect. Soluble fiber in the digestive tract slows the transit of other
materials through the digestive system, resulting in a slower rate of glucose production. Fiber-
rich foods are typically low in fat and added sugar.

1. What is Sexism and how does it show up in My (Your) Life?
Sexism is an act of treating unfairly of oneself because of their sex, mostly it
applies to women. Sexism show to my life as I am a woman myself. Being a woman has
a special treatment, either it is good or bad.
2. When has someone doubted your ability to do something because of your gender?
When I was in high school, I decided to live in a boarding house with my two
cousins in Tacloban City. My parents are hesitant to allow me because they believe I am
incapable of doing so, and my safety is compromised because I am living in a city with
only my cousins. They are also concerned about my health, knowing how easily I catch
illnesses.
3. When has someone assumed you could do something because of your gender?
When my family expects me to do a better job in school because girls perform
better in school than boys. They see it in my mother's siblings, so they expect me, and
us, to be the same. Even if my cousins and I make a joke about not going to school,
most of them get angry and give us a scolding that isn't forgetful. Education is the most
precious thing we have, and we must cherish it and not waste it, as my family says.
4. When have you felt excluded from a conversation, situation, process or decision
because of your gender?
Being excluded is extremely difficult, especially when it is based on gender. I was
once excluded from the family decision-making process. They made a decision on a
very important matter in which my opinions and perceptions were not taken into account
because I was a girl and young at the time.
5. When have you felt shame or inadequacy because you didn’t live up to expectations
placed on you because of your gender?
When we used to have a recitation in one subject. When I was in grade 11, our
adviser was having a discussion when she suddenly called my name, and as a new
student there, I didn't get the chance to answer her question because I was
embarrassed, and my social anxiety kicked in. They anticipate that I will be able to
answer the question because the majority of the girls study and pay close attention to
the teacher.
6. When have you felt uncomfortable or physically intimidated by another person’s body
language, either because of their gender or your gender?

During the pre-interaction phase, I observed him quietly and intently to know his feelings and
emotions regarding to the interview that I will conduct with him. He looks intense and nervous
as I didn’t tell him beforehand the questions that I will ask him, all I ever told him was that the
topic is all about mental health focusing on stress. Yet I give him an assurance that the
interview that I will conduct is confidential and will be a great help for me. I also observed that
he prepares the interview by showing a neat and clean look that is appropriate for the
interview, and introduces himself for formality. And before the interview starts, he started to
calm as he is convinced the assurances that I gave to him.
