Lipids Lab: A. Spotting Effect

The document summarizes several lipid tests performed in a lipids lab: 1. The spotting effect test identifies the presence of lipids like oils, waxes, and fats based on their solubility properties. Vegetable oils are unsaturated and soluble in methylene chloride, while lecithin is insoluble in water. 2. A halogenation test identifies unsaturated lipids by their reaction with halogens to form addition products, visible as decolorization of vegetable oils. 3. The Molisch test identifies glycolipids containing both lipids and carbohydrates through their reaction with furfural to form a visible violet ring. 4. Tests identify the presence of sterols like

Lipids Lab

A. Spotting Effect
- presence of LIPIDS
- ex: oil, wax, fats, etc.
- Fats: Saturated (animals)
- Oil: Unsaturated (Vegtables)
- Carboxylic Acid + long chain of Hydrocarbons = Lipids
- Dimethicone: Vitress Hair Oil
: High Boiling Point, Low Volatility

B. Solubility
-Vegetable Oil: miscible in Methylyn Chloride (Slightly polar, slightly non-polar)
- “Like Dissolves Like”
- Lecithin: immiscible in water
- Lipids: non polar

C. Test for Unsaturation

-presence of DOUBLE BONDS IN UNSATURATED LIPIDS

-principle: Halogenation via Addition

-(+) Visible Result: Decolorization

- Vegetable Oil: Positive

Extraction of Brain Lipids

-Brain + Ether

-Brain: sphingolipids + other lipids in the brain

-Ether: Extracts Lipids

Molisch Test
-Lipids with Carbohydrates (Glycolipids- Galactolipids)

-(+) Visible Result: Violet Ring

-Furfural: reacts with Alpha Naphthol

Soda Lime Test

- Nitrogen containing compound
- Red > Blue = Basic
- Ammonia (Base)
- NH3 + H2O = NIH4 + OH
- reaction by Amino (Ionized) / Imino group

Leitermann-Burchard Test
-Sterol

-Cholesterol: fats present in the brain

-ResidueC: Dark Green (+)

-Lecithin: (+) No sterol (??)

-(+) Visible Result: Green
