Src mediates TGF-β induced Smad1 phosphorylation.

(A) Foreskin fibroblasts were transfected with control (SCR) or Src siRNA oligos. pSmad1 and total Smad1 were analyzed by western blot. (B) Foreskin fibroblasts were transduced with dominant negative Src adenovirus and then stimulated with TGF-β for 30 minutes. pSmad1 and total Smad1 were examined by western blot. (C) Foreskin fibroblasts were transfected with Src siRNA oligos (top panel) or transduced with dominant negative Src adenovirus (lower panel), then stimulated with TGF-β for 24 hours and examined for CCN2 and collagen levels by western blot.
