Preparation
Small beaker and centrifuge tubes were pre chilled in ice bucket. 20 grams of each leaf (river
plant, dry plant and spinach) was measured. For each leaf, the stem was removed and combined
with 80mL of sucrose buffer into a blender. Blended solution is obtained and filtered through a
cheese cloth. 13 mL of filtrate was transferred to a centrifuge tube and centrifuged at 1000 rpm for
10 minutes. Supernatant was obtained and suspended in sucrose buffer. Note that the above
procedure was done for each leaf. Small sample of supernatant was stored in cuvettes and the rest
used for the following steps.

In a tube for each leaf (river plant and dry plant) solution, the following were added; 9 drops of Pi
buffer, 6 drops ADP, 6 drops PMS and 6 drop of the plant s chloroplast solution. The same was
done with two tubes containing spinach. ATP was however added to one tube and PMS was not
added to the other. Each tube was gently mixed and exposed to light for 10 minutes.
